Technology Overview

Pacific Biosystems Sequel

PacBio sequencing is a high throughput / third-generation sequencing technology which is also known as single-molecule real-time (SMRT) sequencing. It utilizes zero-mode waveguides (ZMWs) to capture the real-time incorporation of nucleotides by DNA polymerase. This technology consistently produces long reads (average > 10,000 bp) which can span complex or repetitive regions with a single continuous read to enable high-quality of de novo assembly. It is also good for the survey of full-length transcript isoforms. Our PacBio platform adopted streamlined and consolidated protocols to support multiplexed library preparation and sequencing with different PacBio barcode system, offering cost-effective solutions for multiple applications.

The PacBio Sequel system has been installed at the Core in June 2017. The Core has extensive experience in handling various type of projects, ranging from full-length 16S rRNA sequencing to human whole genome sequencing. Protocols are being actively developed.
